Isaac's Awakening is a new challenge unlocked by beating The Witness for the first time. The challenge tasks you with making it through the Antibirth chapters with the specific setup the game gives you.
At the current date, it does unlock nothing and will remain un-crossed on the challenge list, due to a bug.
